人卵母细胞在不同培养液下成熟与凋亡的比较性研究

为了解卵母细胞体外成熟与凋亡过程，进而提高卵母细胞体丰成熟率，本文采自手术标本中卵巢及卵巢组织，吸取４～１０ｍｍ卵泡，获得未成熟卵母细胞，用正交设计，于培养液中加入不同量组合的人绝经期促性性激素（ｈＭＧ），卵泡液，颗粒细胞，在３７℃，ＣＯ２５％培养箱中培养，并应用形态学观察和原位ＤＮＡ片段末端示记（Ｔｕｎｅｌ）检测卵母细胞的凋亡状况，结果：在本研究体外培养条件下能使未成熟卵母细胞成熟，受精。

To study human oocytes maturation and apoptosis in vitro,and increase rate of oocytes matu-ration, immature oocytes obtained from surgically removed ovaries or ovarian tissue were incubat-ed to mature in Ham's FlO with different concentration of hMG, follicular fluid and granulosa cells by an orthogonal design;morphological examination and Tunel method were employed to de-tect apoptosis of oocytes. Results;Immature oocytes can mature and the mature oocytes can be fertilized .and the fertilized ova can develop into early blastocysts in vitro;The medium consisted of 0. 15 lU/nil hMG,40% follicular fluid and 102/ml granulosa cells gave the highest rate of mat-uration, fertilization and cleavage,and the lowest rate of apoptosis (P<0. 05..P<CO. 005). These results suggest that culture medium consisted of hMG .follicular fluid and granulosa cells in suit-abe concentations can promote immature oocytes to become mature in vitro. and the mature oocytes can be fertilized to reach the early blasocyst stage.
