摘要
采用紫外诱变方法对实验室保藏的粗酶活为10U/mL的野生微球菌SX-1进行诱变处理以获得高产酶能力的菌株,实验得到最佳诱变条件为:15W紫外灯,垂直照射距离为30cm,处理时间为90s,通过荧光圈初筛和摇瓶复筛,筛选出一株突变菌株,酶活可达到22.5U/mL,比出发菌株脂肪酶活力提高了125%,将其连续传代5次,酶活稳定,是一株比较理想的脂肪酶产生菌.
In order to obtain mutants with high lipase production, the Micrococcus SX-1 was taken as initial strain, which enzyme activity was 10 U/ml, and was treated with ultraviolet(UV) radiation. And the optimum treating conditions which was got by primary screening were as follows: the 15 W UV lamp was used as UV light with 30 cm radiation distance and 90 s treating time. Through UV mutation a mutant strain named SU-1 was obtained. As compared with the initial strain, the activity of lipase had reached 22.5 U/mL , which was 125% higher than that of the initial strain. Then an experiment for studying the genetic stability was took by means of passaging 5 times and its results showed that Micrococcus SU-1 had a better genetic stability.
