一氧化氮对人离体多核粒细胞不同状况下呼吸爆发功能的影响 被引量:1

摘要 本研究拟观察不同浓度的一氧化氮(NO)气体及其水溶液对人体离体不同状态下多核粒细胞(PMN)呼吸爆发功能的影响及量-效关系。资料和方法多核粒细胞(PMN)的制备选择30例ASA-Ⅰ级的先天性心脏病行心脏手术的病人,于术前每人取动脉血20ml,肝素钠抗凝(每ml血用肝素钠20U)。采用一步梯度离心沉降法分离人体PMN将所有收集到的PMN加PBS稀释至20ml备用,同时进行细胞计数。 NO气体的制备将NO气体储气瓶(800ppm,氮气平衡,北京氧气厂)与混合气瓶(5%CO_2、95%O_2自行配制)减压后通过微流量表混合。
