原发性膀胱癌多肿瘤抑制基因1改变及其意义 被引量:2

Alteration of multiple tumour suppressor 1 (MTS1) in primary bladder cancer

摘要 为探讨多肿瘤抑制基因1(MTS1)在原发性膀胱癌发生发展中的作用,采用PCRSSCP和DNA直接测序方法,对18例膀胱移行细胞癌进行MTS1分析。结果18例原发性膀胱癌中没有发现MTS1纯合性缺失,但发现2例点突变。 Multiple tumor suppressor 1(MTS1), encoding a Mr16 000 protein (p16) ,maps to chromosomal 9p21. The p16 protein inhibit the kinase activity of cyclin D/CDK4 complex and blocks its pregression to S phage, thus inhibiting cell growth and division and blocks the malignant transformation of cell. When p16 gene was inactivated resulting from homozygous deletion or point mutation, its function as a tumor inhibitor will be lost,resuling in tumor development. To elucidate the role of MTS1 gene in tumorigenesis of bladder cancer,18 cases of bladder cancer were analyzed by polymerase chain reaction(PCR),PCR single strand conformational polymorphism (SSCP) and DNA sequencing. No homozygous deletions were found in the MTS1 gene,while point mutations were found in 2 of 18 bladder cancers. It was concluded that tumorigenesis of primary bladder cancer is correlated with inactivation of MTS1 gene.
