Caspase-3、PARP在四氢异蓰草酮诱导HepG-2细胞凋亡中的变化

摘要 目的研究caspase-3、PARP在四氢异薇草酮诱导HepG-2细胞凋亡中的变化,初步探讨四氢异捧草酮诱导H印G一2细胞凋亡的机制。方法浓度为10、0、40μg/mL的四氢异蓓草酮对HepG一2细胞作用24h后,采用Westernb10t法检测四氢异耩草酮对HepG-2细胞中Cmpme-3、PARP蛋白表达的影响。结果不同浓度的TH对HepG一2细胞作用24h后,HepG一2细胞的Cmpase-3、PARP蛋白表达水平随着TH浓度的升高而升高,并具有良好的量效关系,与阴性对照组比较差异显著(P〈0.05),具有统计学意义。结论TH诱导HepG-2凋亡的机制与上调Caspase-3、PARP蛋白表达有关。 AIM To study the relation between the Caspase -3 and PARP protein and the poptosis of HepG -2 cells induced by Tetrahydre- iso- humulone (TH) . Methods HepG- 2 treated with 10,20,40μg/mL TH for 24h, Caspase -3 and PARP expressions of HepG - 2 cells were detected by Western blotting. ResultS The results showed, TH can rise Caspase - 3 and PARP of HepG -2 ceils after treated with different dosages of TH, TH groups have significant difference compared with negative control group (P 〈 0. 05) and the results have statistical significance. Conclusion Therefore, TH can induce tumor ceils to apoptosis are their main anti - tumor mechanism. Their inducing apoptesis mechanism are related to rising Caspase -3 and PARP protein expression.
