参麦注射液对脑出血大鼠脑组织HSP70表达的影响及其神经保护作用机制的实验研究 被引量:7

摘要 目的:观察参麦注射液对脑出血大鼠血肿中心区、缺血半暗带区、术侧海马区和皮层区HSP70表达的影响。方法:将50只SD大鼠随机分为正常组、假手术组、模型组、参麦组共4组,模型组和参麦组内又分为1d、3d、5d、7d共4个时间点,立体定位仪定位注射胶原酶VII造大鼠脑出血模型。评估各组大鼠神经系统病态,并进行爬杆计分,电镜观察脑出血后的神经细胞的病理改变,免疫组织化学法检测各组大鼠脑出血后不同时间点血肿中心区、血肿周围区、术侧海马区及皮层区神经细胞HSP70的表达,以及参麦注射液对其的影响。结果:正常组及假手术组大鼠未见明显的病态,模型组和参麦组均出现不同程度的病态体征;电镜观察显示,正常组和假手术组神经细胞结构正常,模型组细胞器不完整,出现了凋亡小体,而参麦组细胞器较完整,凋亡小体较少;正常组和假手术组大鼠海马区和皮层区神经细胞有少量的HSP70阳性信号表达,模型组缺血半暗带区、术侧海马区和皮层区HSP70的表达1d时增多,3d时达到高峰,5d时表达下降,至7d时仍有少量表达,与模型组相比较,参麦组各时间点的HSP70表达增多,差异有统计学意义(P<0.01)。结论:参麦注射液可能通过增加HSP70的表达,抑制细胞凋亡,增加神经细胞耐缺氧能力而发挥神经保护作用。 Objective:To observe the effects of ShenMai Injection on the expression of the heat shock proteins70(HSP70) in the areas of hematoma,ischamic Penumbra(IP),hippocampal gyrus and cotex of operated sides.Methods:The 50 rats were randomized into 4 groups:normal groups,false operation groups,modeling groups,and treating groups with Shenmai Injection(SM groups),the rats in the modeling groups and treating groups with Shenmai were made into four stages respectively in 1d,3d,5d,7d after operation,the models of Intracerebral Hemorrhage were made by injecting Collagenase type Ⅶ stereotaxically;we evaluated the pathosis of all the rats,and counted the scores of climbing haulm,the pathological changes of neurocyte were observed by electron microscope.Subsquently,we observed the expression of heat shock proteins 70 in the perihematomal area by immunohistochem at the different stages in the the areas of hematoma,IP,hippocampal gyrus and cotex of operated sides,and the effect of Shenmai Injecton on them.Result:Rats in the normal groups and false groups indicated no pathological signs.However,Rats in the modeling groups and SM groups desmonstrated obvious pathological signs.The observation by the electron microscope displayed that the rat,s cytoarchitecture in the normal groups and false groups is normal.that the cytoarchitecture in the modeling groups is not integrated,the apoptosis bodies found in their cytoarchitecture were more than them in the SM groups.A few of expression of HSP70 can be observed in the rats of normal groups and false operation groups.However,the expression of HSP70 increased significantly in the areas of IP,hippocampal gyrus and cotex of operated sides,got to a peak at 3st day,followly decreased gradually,kept higher at 7st day.SM Injection can enhance the levels of the expression of HSP70.there were significant difference between modeling groups and SM groups.Conclusion:SM Injection can promote expression of HSP70,which can inhibt neuronal apoptosis,and increase the ability of neuronal tolerance to hypoxia.provide a protctive contribution.
