摘要
研究了用无载体的188Re直接标记单克隆抗体的方法,以摩尔比为1∶3的柠檬酸-酒石酸作中间螯合剂,在pH=5.2的条件下,用SnCl2将Na188ReO4快速还原,再与已被NaHSO3还原的抗体交换配位。研究结果表明,反应2h的标记率大于95%,标记物的体内、体外稳定性良好并保持了抗体的免疫活性。
A convenient and efficient method of direct labeling monoclonal antibodies with 188 Re is established. The rapid and complete reduction of 188 ReO 4 is achieved with SnCl 2 in an aqueous mixture solution of citric acid and tartaric acid (molar ratio=1∶3) at pH 5.2. The reduced 188 Re solution is then mixed with a NaHSO 3 - reduced antibody solution and incubated at room temperature for 2h. The mean labeling efficiency is higher than 95%.The labeled antibody i found stable in vivo and in vitro, and is immunoactive.
