乙型肝炎病毒相关性肝癌患者的蛋白质组学研究 被引量:1

Proteomic Study of Differential Protein Expression in HBV-related Primary Hepatic Carcinoma

摘要 目的初步探讨蛋白质组技术在精浆研究中的应用,建立乙型肝炎病毒相关性肝癌及健康者血清双向凝胶电泳图谱;筛查并鉴定二者间的差异表达蛋白质,期望从中发现有意义的分子标志物。方法采用双向凝胶电泳(2-DE)分离乙型肝炎病毒相关性肝癌和健康者血清总蛋白,银染显色,PDQuest7.3.0软件分析,从中选取差异表达蛋白质,通过基质辅助激光解析电离飞行时间质谱(MALDI-TOF-MS)鉴定差异表达的蛋白质。结果获得分辨率较好的精浆双向凝胶电泳图谱;得到8个在乙型肝炎病毒相关性肝癌与健康者血清中差异有统计学意义的蛋白质斑点,进行鉴定,结果表明这些特异蛋白分别为肝脂肪酸结合蛋白、谷胱甘肽S转移酶A1、27×103热休克蛋白、黄素还原酶、假定蛋白、醛酮还原酶、乙二醛酶、细胞角蛋白18。结论在乙型肝炎病毒相关性肝癌与健康者血清中表达蛋白质差异有统计学意义,利用蛋白质组学这一高通量的筛查技术可望从中发现与乙型肝炎病毒相关性肝癌相关的血清标志物。 OBJECTIVE To investigate the application of proteomics in the field of serology,and to screen the differential expression proteins associated with HBV-related primary hepatic carcinoma. METHODS Two-dimensional electrophoresis was applied to segregate the total proteins in the serum from patients with HBV-related primary hepatic carcinoma and health volunteers. After staining,the differential expression proteins were analyzed using PDQuest7.3.0 software,and identified using matrix-assisted laser desorption/ionization-time of flight mass spectrometry (MALDI-TOF-MS). RESULTS Electrophoresis figures with high reproducibility were obtained. The eight high-abundance protein spots were identified and analyzed. The special protein spots were liver fatty acid binding protein,glutathione S-transfer A1,27×10^3 heat-shock protein,flavin reductase,hypothetical protein,aldehydes and ketones reductase,GLO,and CK18. CONCLUSIONS Protein expression is differential in the serum from the patients with HBV-related primary hepatic carcinoma and health volunteers. It is hopeful to find the biomarkers related with poorly differentiated hepatic carcinoma using proteomics.
