RhoA/Rho激酶信号通路对小鼠抗衰老Klotho蛋白表达的影响 被引量:6

Effect of RhoA/Rho-kinase on the expression of anti-aging klotho protein in the brain of mouse

摘要 目的观测RhoA/ROCK信号通路对小鼠脑脉络丛Klotho(KL)蛋白表达的影响。方法将24只昆明小鼠随机分为3组,即对照组,一氧化氮合酶(Nitric Oxide Synthesis,NOS)抑制组(L-NAME组)和L-NAME+法舒地尔组,连续处理4周。用免疫组织化学方法观察KL蛋白表达;Western blot检测KL蛋白表达水平的变化;RT-PCR检测RhoA和ROCK mRNA表达水平。结果与对照组比较,L-NAME组ROCK mRNA表达水平升高(P<0.01),KL蛋白表达水平明显降低(P<0.01)。给予ROCK阻滞剂法舒地尔后ROCK mRNA表达水平降低(P<0.01),Kl蛋白表达水平明显增高(P<0.01)。结论L-NAME所致KL蛋白表达下调与RhoA/ROCK信号通路活性增强有关。 Objective To investigate the effect of RhoA/Rho-kinase pathway on the expression of Klotho(KL) protein in the brain choroid plexus in mouse.Methods 24 male Kunming mice were divided randomly into three groups: the normal control group,NOS blockade(L-NAME) group,and L-NAME plus fasudil group,and were administered by corresponding medicine respectively,for four weeks successively.The expression of KL protein in the brain choroid plexus were measured by immunohistochemistry and Western blotting.The expression of RhoA/Rho-kinase mRNA were detected by RT-PCR.Results Compared with the control group,the expression of RhoA/Rho-kinase mRNA increased significantly(P〈0.01) and the expression of KL protein decreased sharply(P〈0.01) in the L-NAME group.Fasudil can reverse these changes(P〈0.01).Conclusions These results suggest that L-NAME could induced downregulation of expression of KL protein via activated RhoA/Rho-kinase pathway in mouse brain.
