摘要
目的探讨颞叶癫痫大鼠海马组织中miRNA分子表达谱的差异,为进一步研究相关miRNA在颞叶癫痫发病机制中的作用打下基础。方法对同一父系和母系的子代大鼠,利用氯化锂-匹罗卡品化学诱导方法制备慢性颞叶癫痈大鼠模型。分别提取1只正常和3只颞叶癫痫大鼠海马组织的miRNA,采用高通量的miRNA微阵列芯片杂交,筛选颞叶癫痫海马组织中差异表达的内源性miRNA。结果在大鼠海马组织中共检测到125个miRNA基因。与正常大鼠相比,颞叶癫痫大鼠海马组织中差异表达的miRNA有23个,其中有5个miRNA下调,18个miRNA上调。结论与正常大鼠相比,颞叶癫痫大鼠海马组织中存在差异表达的miRNA分子,差异表达的miRNA分子可能参与癫痫的发病过程,具有潜在的研究价值。
Objective To explore the difference of miRNA expression in hippocampus in temporal lobe epilepsy (TLE) for further studies on the functions of miRNA in the pathogenesis of temporal lobe epilepsy. Methods In the offsprings of the same maternal and paternal rats, established the rat models of chronic TLE by lithium-pilocarpine induction. Total miRNA was isolated from the hippocampus of one normal and three chronic TLE rats. High flux miRNA microarray chip was used to screen the expression miRNAs profile whose differential expression is greater than 50% between groups of TLE rats and the control in the hippocampus. Results All 125 effective miRNAs expression data were obtained in the hippocampus and among them, 23 miRNAs expression level had greater than 50% difference in TLE group compared with control rats, including a set of 5 under-expressed miRNAs and 18 over-expressed miRNAs. Conclusions There are different expression miRNAs between chronic TLE model and the normal rat. These differential expression miRNAs may be involved in the pathogenesis of epilepsy and worth further study.
