摘要
目的研究癫痫患者CYP2C19基因多态性。方法运用PCR-RFLP方法对67例应用丙戊酸的癫痫患者的CYP2C19*2和CYP2C19*3位点进行基因型分析。结果两个位点共有5种基因型组合:681GG-636GG、681GA-636GG、681GG-636GA、681AA-636GG、681GA-636GA。快代谢者(EMs)包括681GG-636GG、681GA-636GG、681GG-636GA基因型,分布频率为86.57%;慢代谢者(PMs)包括681AA-636GG、681GA-636GA基因型,分布频率为13.43%。结论PMs发生率与国内报道相近,CYP2C19基因多态性分析有助于丙戊酸的临床合理应用。
Objective To investigate the genetic polymorphism of CYP2Ct9 in patients with epilepsy. Methods CYP2C19 variants ( * 2 and * 3 ) were detected by PCR-RFLP method in 67 patients taking valproic acid. Results We found that there were five genotypes of CYP2C19 in these subjects,and that the frequencies of Extensive Metabolizers (EMs)were 86. 57% and the frequencies of Poor Metabolizers (PMs)were 13.43%. Conclusion The frequencies of PMs are similar to the data of the report. The genetic polymorphism analysis of CYP2C19 is helpful for the reasonable use of valproic acid.
