冠心Ⅱ号不同组分配伍对大鼠急性心肌梗死的影响

Effects of Different Compositions of Guanxin Ⅱ(GXⅡ)Prescription on Experimental Myocardial Infarct in Rats

目的观察冠心Ⅱ号全方中活血药、理气药以及二者配伍对大鼠急性心肌梗死的影响,探讨冠心Ⅱ号配伍规律。方法冠状动脉结扎法制备大鼠急性心肌梗死模型;造模后5～10min十二指肠给药;造模后5h,腹主动脉取血,测定血浆中内皮素(ET)和血栓素B(2TXB2)水平,血清肌酸激酶同工酶MB(CK-MB)和乳酸脱氢酶(LDH)水平;取血完毕后,取出心脏,切片、硝基四氮唑蓝(N-BT)染色、扫描,测定心肌梗死面积。结果冠心Ⅱ号全方组和活血药组的心肌梗死面积、血浆ET和TXB2水平、血清CK-MB和LDH水平均显著下降(P<0.01或P<0.05),二者作用强度无显著性差异;而理气药组对心肌梗死面积、血浆ET和TXB2水平无显著影响,仅对血清CK-MB和LDH水平有显著作用。结论冠心Ⅱ号全方中的活血药在抑制心肌缺血作用中发挥了主要作用,而理气药起到一定的辅助作用。

Objective To compare effects of the whole prescription of Guanxin II （GXⅡ）, the hemorheologic agents and the Qi-regulating agents of the whole prescription on acute myocardial infarction in rats. Methods Acute my- ocardial infarct （AMI）model was established by coronary artery ligation, and then the model rats were treated with in- traduodenal administration of different compositions of GXII 5 -10 minutes after the artery ligation. Five hours after modeling, blood was collected to determine the levels of endothelin（ET）, thromboxane（TXB2）, isoenzymes of crea- tine kinase （CK-MB）and andlactate dehydrogenase （LDH）. The heart of rats was cut into five slices and dyed by ni- trotetrazolium blue chloride（N-BT）for 5 minutes, and then the infarcted area（non-N-BT staining area）and the non- infarcted area（N-BT staining area）were detected. Results Compared to the model group, both the whole prescription of GXII and the hemorheologic agents significantly reduced infarct size, ameliorated blood ET, TXB2, CK-MB and LDH levels （P 〈 0.05 or P 〈 0.01）, and they had the similar efficacy. However, the Qi-regulating agents of the whole prescription did not show any effect on infarct size and blood levels of ET and TXB2, but ameliorated blood levels of CK-MB and LDH. Conclusion In the whole prescription of GXⅡ, it is the hemorheologic agents which mostly contribute to the effect on inhibiting myocardial ischemia, and the Qi-regulating agents only shows certain see- ondary effect.
