摘要
目的研究在慢性酒精中毒性肌病中的线粒体损伤情况并探讨受损原因。方法建立大鼠慢性酒精中毒性肌病模型,采用分光光度计法、Western Blot和流式细胞仪检测实验组大鼠骨骼肌各抗氧化指标(SOD、GSH-Px、MDA)、线粒体膜蛋白细胞色素C、线粒体膜流动性和线粒体膜电位的变化。结果实验组大鼠骨骼肌中MDA含量升高,SOD、GSH-Px活性降低,细胞色素C的表达减少,线粒体膜流动性下降,线粒体膜电位降低。结论慢性酒精中毒性肌病中线粒体发生损伤,提示与活性氧(ROS)增多有关。
Objective To study mtDNA lesions in the chronic alcoholic myopathy and to explore the reason of the lesions.Methods Spectrophotometer,western blotting and flow cytometry analysis were utilized to determine the SOD、GSH-Px and MDA,the expression of cyt-C,mitochondrial membrane fluidity and mitochondrial membrane potential(△Ψm) in the experimental group.Results MDA were significantly higher,activity of SOD and GSH were lower,mitochondrial membrane fluidity were lower,the expression of cyt-C decrease and mitochondrial membrane potential were lower in the experimental group.Conclusion The mtDNA lesions in the CAM relates to the increase of ROS.
