摘要
目的:探讨低氧性肺动脉高压大鼠肺组织Ⅰ、Ⅲ型胶原的变化及其与肺动脉压的关系。方法:经右心导管法检测肺动脉压和肺循环阻力等血流动力学指标,以链亲合素过氧化物酶法观察缺氧大鼠肺动脉、支气管和肺间质Ⅰ型和Ⅲ型胶原的分布变化。用图像分析技术检测肺小动脉的形态改变和胶原组化染色灰度扫描。结果:缺氧3周大鼠肺动脉压明显升高,Ⅰ、Ⅲ型胶原表达均明显增加,Ⅰ型胶原主要分布在肺动脉壁外膜,Ⅲ型胶原主要分布在内膜。结论:肺动壁Ⅰ、Ⅲ型胶原增多与肺动脉高压的形成密切相关。
Object: To investigate the changes of type I and Ⅲ collagens in the lung tissues of rats with hypoxic pulmonary hypertension (HPH) and its relationship to the mean pulmonary arterial pressure (mPAP). Methods: The blood dynamic idexes of pulmonary circulation were measured by using Swan - Ganz, The distribution of type I and ± collagens in pulmonary arteries, bronchi and pulmonary interstitial was observed by using streptavidin peroxidase method (SP) the morphologic changes of small pulmonary arteries and the level of type I and Ⅲ collagens were determined by image pattern analysis technique and gray scale scanning respectively. Results: Compared with the control group in the hypoxic rats the mPAP elevated and the collagens of type I and III increased remarkably, futhermore, the distribution of type I collagen was mainly in the external layer of pulmonary arterial walls, but type Ⅲ collagen mainly in the internal layer. Conclusion: The increase of type I and Ⅲ collagen contents in pulmonary arteries is closely related to the development of HPH.
