延庆地区侵染百合的病毒检测与序列分析 被引量:1

Detection and sequence analysis of viral disease in infected lily from Beijing

摘要 利用DAS-ELISA和RT-PCR等检测方法,对北京市延庆地区疑似感染病毒的百合植株进行了检测,并将RT-PCR扩增到的产物进行克隆及序列分析验证。DAS-ELISA结果表明:百合叶片的粗汁液与TuBV的抗体呈现阳性,初步确定北京市延庆地区百合感染郁金香杂色病毒(TuBV)。RT-PCR检测结合克隆测序结果表明:郁金香碎色病毒CP基因的核苷酸序列与GenBank上已登录的(AB090385.1,AB078007.1和S44147.1)郁金香碎色病毒的核苷酸序列的同源性均为100%,与已登录的(qb|AAB19407.1|,dbj|BAD02938.1|和dbj|BAB83899.1|)氨基酸序列同源性均在95%以上,同源性较高,进一步证明北京市延庆地区百合感染有郁金香杂色病毒(TuBV)。 Virus species infected lily were detected by using DAS-ELISA,RT-PCR and nucleotide(nt) sequence analysis of coat protein(CP) gene.The result of DAS-ELISA revealed that rough juice of lily was serologically related to Tulip breaking virus.It was determined preliminarily that Tulip breaking virus(TuBV) was the virus infecting lily in Yanqing Beijing.The CP gene of TuBV from lily was cloned and sequenced.The sequence analysis showed that the nucleotide(nt) and aminoacid(aa) sequence of TuBV from lily CP genes(GenBank accession No.: AB090385.1,AB078007.1,S44147.1 and qb|AAB19407.1|,dbj|BAD02938.1|,dbj|BAB83899.1|)were 100% and 95% identical to other TuBV strains.According to the sequence analysis of coat protein gene,we concluded that the lily in Yanqing Beijing were infected by TuBV.
