细胞因子对人绒毛瘤细胞JARβ-HCG分泌的影响

摘要 人绒毛股促性腺激素(HCG)的分泌调节机制至今仍不详。本文以人绒毛瘤细胞JAR作为人滋养层细胞的模型,研究了IFNα、TNF-α、M-CSF、TGF-β、UK和NGF等细胞因子对β-HCG分泌的影响。结果显示,IFNα和TNF-α。能显著地促进β-HCG的分泌,并具有剂量依赖性,UK则能显著地抑制β-HCG的分泌,而M-CSF、TGF-β和NGF则无明显作用。细胞活性实验表明.高浓度的IFNα、TNF-α和TGF-β能显著抑制细胞的生长,而其他细胞因子则无作用。以上结果提示,细胞因子可以通过影响人滋养层细胞的生长和分泌而成为其功能活动的重要调节因素。 The regulation of human chorionic gonadotropin secretion is not fully understood. The present study was conducted to investigate the effects of cytokines on secretion of beta-HCG in human trophoblast cell, JAR. The results revealed that IFN ot and TNF- α significantly stimulated β-HCG re1ease in a dose-dependent mauner. However, UK inhibited β - bCG secretiou significantly. The othAr cytokines had no significant effects. The study on cell viability revealed that IFN α, TNF- α and TGF-β inhibited the growtb of JAR, but theotbers didnt. These results suggest tbat cytokines may bA important regulators of trophoblast function in influencing tbe growth and secretion of buman trophoblast cells.
