摘要
应用细胞培养技术对新生牛主动脉平滑肌细胞进行传代培养,取第4~6代平滑肌细胞作为受试对象研究不同浓度胰岛素对牛生动脉平论肌细胞内脂质含量的影响,采用Heider改良法(即酶-荧光法)测定平滑肌细胞内微量胆固醇。结果显示:胰岛素增加平滑肌细胞内胆固醇的含量,且胰岛素浓度与平滑肌细胞内胆固醇含量是正相关。上述结果提示:胰岛素可促进平滑肌细胞内脂质蓄积,从另一侧面加速动脉粥样硬化的发生与发展。
By using the cell culture technique, sub-cultures of bovine aortic smooth muscle cells were proceeded. We investigated the effects of different concentrations of insulin on the anlount of (tholestprol in smooth muscle cells (passage4 ~ 6). Ameliorative Heider's method (an enzymatic fluoromatric method) was applied to the determination of either total or free cholesterol in smooth muscle cells The results showed that: insulin could markedly increase cholesterol (fred cholesterol,total cholesterol and cholesteryl ester) contents in smooth muscle cells: there were significant positive correlations between insulin concentrations and cholesterol contents in smooth muscle cells.The results suggest that insulin may contribute to the development of the lesions of atherosclerosis by promoting accumulations of lipid in smooth muscle cells.
