摘要
本文分析了影响共焦荧光扫描显微镜的各种因素——探测器小孔直径、杂散光、物镜孔径等,并以正在研制的一台共焦荧光扫描显微镜为例,讨论了如何抑制杂散光,在设计上保证系统高分辨率实现的方法。
This paper discusses various factors that influence the resolution of a confocal fluorescent microscope,including the diameter of the pinhole in front of the detector,stray light and the numerical aperture.A design method is also presented in order to ensure the realization of the super resolution of the system.The images of thin optical sections in thick samples are given which show that the instrument designed with the method makes possible a new type of noninvasive optical biospy.
