依托考昔对低氧缺糖诱导PC12细胞损伤的保护作用 被引量:2

摘要 目的:探讨环氧化酶-2(COX-2)抑制剂依托考昔对PC12细胞低氧缺糖损伤(OGD)的保护作用。方法:以PC12细胞OGD损伤模拟脑缺血损伤模型,依托考昔的给药浓度分别为10、20、40、80、160mmol/L,采用光镜观察细胞形态学变化,以MTT法检测细胞存活率,乳酸脱氢酶(LDH)法检测细胞损伤。结果:依托考昔(40~80mmol/L)能增加OGD损伤后PC12细胞的存活率,并减少LDH释放(P<0.05)。结论:COX-2抑制剂依托考昔对OGD诱导的PC12细胞损伤具有保护作用。 Objective: To investigate the protective effect of COX-2 inhibitor Etoricoxib on oxygen glucose deprivation (OGD)-induced injuries in PC12 cells. Methods: PC12 cells were exposed to OGD, with or without Etoricoxib at different doses (10, 20, 40, 80, 160 mmol/L). The morphology of cells was detected by light microscope, the cell viability and injury were detected by MTT assay and the lactate dehydrogenase (LDH) kit, and the expression of COX-2 in cells was detected by immunochemistry. Results: After treated with Etoricoxib, the cell viability was increased obviously at the concentration of 40-80 mmol/L, and the efflux of LDH was reduced significantly (P〈0.01). Conclusion: COX-2 inhibitor Etoricoxib pro- tects PC12 cells from OGD insult.
