摘要
目的观察不同参数对超声波逆转肿瘤细胞多药耐药效果的影响,筛选出最优化参数组合。方法以不同频率、脉冲和声强的超声波照射亲本和耐药的人肝癌细胞株HepG2和HepG2/R,通过流式细胞术测定细胞对罗丹明123的摄入量,间接反应超声波逆转耐药的效能。结果在选用的0.4、0.6、0.8、1.7MHz 4种频率中,0.8MHz的超声波逆药的效果优于其它3种频率。选用了10%、20%、30%、40%、50%、60%、70%、80%、90%、100%10种脉冲波,以60%的脉冲波效果最佳。在不同的声强条件下,较高声强具有更好的逆转耐药的效果。结论超声波逆转肿瘤细胞多药耐药的效果与频率和脉冲之间不呈简单的直线对应关系,而是存在着最佳频率和最佳脉冲。对HepG2细胞而言,0.8MHz,60%脉冲比为最佳参数组合。较高声强具有较好的逆转耐药的效果。
Objective To observe the influence of different parameters of ultrasound on its reversal effect in tumor multi-drug resistance,and select the optimal acoustic parameters.Methods A variety of frequency,duty cycle and acoustic intensity were employed to radiatc human hcpatoccllular ccll linc HcpG2 and its multi-durg resistant subline HepG2/R.Intake of Rhodamine 123 was determined by flow cytometry to evaluate the reversal effieiency of ultrasound on HepG2/R.Results Totally 4 kinds of frequencies were used,namely 0.4,0.6,0.8 and 1.7MHz among which 0.6MHz demonstrated the best result.Among a series of duty cycles(10%、20%、30%、40%、50%、60%、70%、80%、90%、100%),60% duty cycle exhibited a better reversal effect.Higher acoustic intensity worked better than did the lower ones.Conclusion The reversal efficacy of ultrasound on multi-drug resistant tumors cells does not correlate with frequency and duty cycle in a linear model.There are best frequency and duty cycle correspond to the ideal reversal effect.With regard to HepG2 cell line,0.8MHz frequency and 60% duty cycle are the optimal parameters.As to acoustic intensity,we recommend the higher one.
