慢性重型乙型肝炎患者外周血树突状细胞表型和功能的研究

The study of epitope and function of peripheral blood dendritic cells from chronic hepatitis gravis

目的:研究慢性重型乙型肝炎患者外周血树突状细胞(DC)表型、免疫功能和分泌细胞因子的特性。方法:从20例慢性重型乙型肝炎患者和10例健康人外周血中分离单个核细胞培养DC,用流式细胞仪检测DC的表面标志CD80和CD86的表达。用3H-TdR掺入法检测DC诱导混合性淋巴细胞反应(MLR)的能力。并用ELISA方法检测MLR中细胞因子IL-12的分泌水平。结果:慢性重型乙型肝炎患者组的DC细胞CD80、CD86的表达率较正常人组高(P<0.05),诱导MLR的能力也较强(P<0.05),MLR中细胞因子IL-12的分泌水平也较高(P<0.05)。结论:慢性重型乙型肝炎患者外周血树突状细胞免疫功能亢进,免疫刺激能力也较高。

Objective：To study the epitope,immune function and cytokine of peripheral blood dendritic cells（DC）from chronic hepatitis gravis.Method：PBMC were isolated from 20 patients with chronic hepatitis gravis and 10 healthy individuals,then the DCs were cultured.The surface markers CD80 and CD86 were detected by flow cytometry.The stimulatory capacity was assessed by mixed lymphocyte reaction（MLR）combined with 3H-TdR incorporation.And the levels of IL-12 in the culture supernatants from MLR were measured by enzyme-linked immunosorbent assay（ELISA）.Result：The levels of CD80 and CD86 expressed on DCs,and the capacity of MLR induction by DCs as well as the level of IL-12 were higher in chronic hepatitis gravis group than those in healthy individuals group（P0.05）.Conclusion：The immune function of peripheral blood DC in chronic hepatitis gravis is enhanced,and the stimulatory capacity is strong.
