三氧化二砷诱导胃癌细胞凋亡和Bcl-2表型变化的意义 被引量:3

摘要 目的:探讨三氧化二砷(As2O3)是否能够诱导SGC7901胃癌细胞凋亡,以及Bcl-2的表型变化是否在其中发挥了作用。方法:将不同浓度的三氧化二砷作用于SGC7901细胞,采用CCK-8进行细胞毒检测;采用DAPI染色进行细胞核形态分析;采用免疫荧光显微镜观察不同表型的Bcl-2表达的变化;采用Western Blot蛋白印迹法检测Bcl-2的表达。结果:三氧化二砷可以诱导SGC7901细胞凋亡。三氧化二砷诱导SGC7901细胞的凋亡伴随着Bcl-2表型的变化,且Bcl-2的表达无改变。结论:三氧化二砷可能通过Bcl-2表型的变化诱导SGC7901胃癌细胞凋亡,而不是通过下调Bcl-2的表达。 Objective:To explone the effects of arsenic trioxide(As2O3) on induction of apoptosis in SGC7901 human gastric cancer cells and the phenotype change of Bcl-2.Methods:SGC7901 human gastric cancer cells were treated with different concentrations of arsenic trioxide.Cytotoxicity was measured by using the Cell Counting Assay Kit-8.Analysis of nuclear morphology was observed with DAPI staining and the phenotype change of Bcl-2 was observed under the immunofluorescence microscopy.The expression of Bcl-2 was examined with Western Blot analysis.Results:Arsenic trioxide could induce apoptosis in SGC7901 human gastric cancer cells,and the apoptosis was accompanied with phenotype change of Bcl-2,but arsenic trioxide did not affect expression of Bcl-2.Conclusion:Arsenic in SGC7901 cells induced by arsenic trioxide may be expressed through the induction of Bcl-2 phenotpe change rather than down-regulation of Bcl-2.
