血浆中非蛋白结合疏基化合物光度法测定研究

摘要 用锌粉——盐酸羟胺将胱氨酸和高胱氨酸还原为半胱氨酸和高半胱氨酸,然后在磷酸缓冲介质中与5,5′——二硫双(2——硝基苯甲酸)简称(DTNB)反应,生成黄色化合物,在其最大吸收波长410nm处测产物的吸光度值,从而对巯基化合物总浓度进行定量测定。该方法的精密度、稳定性好、准确度和灵敏度高。用于血浆中非蛋白结合巯基化合物总浓度的测定。 The total concentration of sulphydryls has been determined quantitatively by measure the absorbance of a yellow compound at max=410mm,the yellow compound are obtained through reaction of cysteine and homocysteine (product of cystine and homocystine reduced by zinc power -hydroxylammonium chloride) with 5,5′-dithiobis (2-nitrobenzoicacid) (DTNB) in NaH 2PO 4-Na 2HPO 4 (PH8.4) buffer solutions. With good precision and stability,high accuracy and sensitivity,this method has been used to determine the total concentration of non-protein sulphydryls in plasma. the results are satisfied.
