摘要
研究目的:观察低氧、低氧运动对骨骼肌肌节内骨架蛋白Titin(肌联蛋白)和Nebulin(伴肌动蛋白)含量的影响。研究方法:以雄性SD大鼠为研究对象,分为常氧对照组、常氧运动组、低氧对照组和低氧运动组,进行为期四周的实验,用免疫印迹法分别检测各组各周腓肠肌Titin和Nebulin蛋白含量。研究结果显示,低氧和低氧运动早期均可导致骨骼肌Titin含量的显著减少(P<0.05),对Nebulin的影响不显著(P>0.05),但呈现降低-升高-再降低-再升高的趋势。结论:低氧和低氧运动早期均可引起肌节骨架蛋白Titin的损伤或降解,可能是由于激活了calpain所致。后期Titin和Nebulin均恢复或稍增加。
The purpose of this study was to observe the changes of content of titin and nebulin in skeletal muscle after hypoxia and hypoxic exercise.Male SD rats were randomly divided into four groups: normoxia control group,hypoxia control group,normoxic exercise group,hypoxic exercise group.After four weeks' experiment,The protein content of titin and nebulin of skeletal muscle was detected by western blot.The results showed that the content of titin in skeletal muscle decreased markedly in the beginning of hypoxia and hypoxic exercise,and that the content of nebulin changed little.The results concluded that the damage and degradation of titin happened in the beginning of hypoxia and hypoxic exercise,and gradually recovered.
