基因微阵列技术筛选结肠癌转移相关基因的研究

摘要 目的以cDNA表达基因芯片技术筛选与原发性结肠癌转移相关的基因,对部分基因进行分析,探讨原发性结肠癌转移发生发展的分子机制。方法收集2例未发生转移及2例已发生转移的原发性结肠癌患者的新鲜冻存癌组织。杂交芯片采用含16k余个点的cDNA表达谱芯片。差异表达基因的筛选标准为该基因在50%以上样本中的荧光强度比(ratio比值)>=2或<=0.5。结果基因芯片筛选获得与细胞粘连、细胞周期、信号传导、分子结构等功能相关的差异表达基因共196条,其中上调基因112条,下调基因84条。结论运用cDNA芯片进行原发性结肠癌转移倾向相关基因的表达谱分析,有助于从分子水平全方位阐明结肠癌转移的发生机制及生物学特性。 Objective To identify cancer metastasis related genes in colon cancer and to analyze part of them by cDNA microarray analysis,and to explore its molecular mechanism.Methods Two samples of colon cancer tissues of non-metastasis and 2 of metastasis were taken and freshly frozen respectively.cDNA microarray with more than 16k genes was used.Genes were considered to be up-or down-regulated when the fluorescent intensity ratio was over 2-fold in over 50% of the samples(P0.05).Results Compared with those of non-metastatic colon cancer tissues,there were a total of 112 genes up-regulated and 84 genes down-regulated in already metastatic colon cancer tissues.The function of these genes refered to cell binding,cell cycle,transduction,cell structural-moleculor,and so on.Conclusion Gene expression profiling by cDNA microarray analysis provides molecular understanding of biological properties of cancer.
