不同方式的低能量激光照射对MG-63矿化组织特异性蛋白的mRNA表达的影响及意义

摘要 目的观察不同方式的低能量激光照射(low level laser irradiation,LLLI)对MG-63矿化组织特异性蛋白的mRNA表达的影响及意义。方法将对数生长期的人成骨样细胞MG63随机分为3组,采用Ga-Al-As半导体激光器(808 nm,66 mW)进行照射,照射组1为照射3天,每天照射剂量为1.25 J/cm2,照射组2为照射1天,照射剂量为3.75 J/cm2,两组的照射总剂量均为3.75 J/cm2;对照组不进行激光照射。照射后72 h,利用半定量逆转录聚合酶链式反应(reverse transcriptase polymerase chain reaction,RT-PCR)的方法检测骨桥蛋白(osteopontin,OPN),Ⅰ型胶原蛋白(collagen Ⅰ,Coll Ⅰ)的mRNA的表达量。结果照射组1和照射组2的OPN,Coll Ⅰ的mRNA的表达量均明显高于对照组,有显著性差异(P<0.05);照射组1和照射组2之间没有显著性差异(P>0.05)。结论总剂量为3.75 J/cm2的低能量激光照射可以促进MG-63表达矿化组织特异性蛋白OPN和Coll Ⅰ,可能促进成骨。在相同的照射剂量下,多次照射和单次照射没有明显区别。 Objective To investigate the effect of low level laser irradiation(LLLI)with different methods on expression of mRNA of specific proteins of MG-63 in mineralized tissue.Methods Human osteoblast-like cells MG-63 were irradiated by Ga-Al-As diode laser(808 nm,66 mW) with total dose of 3.75 J/cm2: laser group 1 was 3-day irradiation with 1.25 J/cm2 once daily and laser group 2 was single irradiation with 3.75 J/cm2 on the first day.Untreated cells were used as controls.After 72 h,the expression of osteopontin(OPN) and collagen Ⅰ(Coll Ⅰ)were measured by semi-quantative reverse transcriptase polymerase chain reaction(RT-PCR).Results The expressions of mRNA of OPN and Coll Ⅰ of laser group 1 and laser group 2 were higher than those of the control group with significant difference(P0.05).Moreover,there was no significant difference of the indexes between laser group 1 and laser group 2(P0.05).Conclusion LLLI of 3.75 J/cm2 could enhance the expression of specific proteins of MG-63 in mineralized tissue which may improve bone regeneration.Moreover,under the same dose of LLLI,there is no difference between multiple irradiation and single irradiation.
