外源性骨髓间充质干细胞在肾脏的分布 被引量:1

摘要 目的探讨外源性骨髓间充质干细胞(mesenchymal stem cells,MSCs)在正常肾脏组织的分布,并分析其可能机制。方法采用贴壁离心培养法分离MSCs,并用PKH67标记。将Wistar大鼠随机分为正常组(A组)、干细胞正常移植组(B组)和干细胞照射移植组1(C1组),干细胞照射移植组2(C2组)。A、B、C1、C2组各5只大鼠。分别在移植后的第3、5、7、14天处死大鼠,留取血、尿标本,观察血肌酐(Cr)和尿素氮(BUN)及24 h尿蛋白定量水平。荧光显微镜下观察移植的MSCs在肾脏组织的分布。结果C组与A组比较,移植后的第3、5、7、14天血Cr、BUN及24 h尿蛋白定量水平均无统计学差异(P>0.05);在C组,体外培养的MSCs在移植后的第5天定位于肾脏,主要是肾小管,且与输注方式无关。结论移植MSCs能够迁移、定居于正常肾脏组织并且主要分布于肾小管。 Objective To identify the disposition and the potential mechanism of exogenous bone marrow mesenchymal stem cells(MSCs) in the normal kidneys.Methods Bone marrow MSCs were collected from femurs and tibias of female Wistar mice,then isolated by percoll density gradient centrifugation and labeled with PKH26.Wistar rats were divided randomly into normal group(A),transplanted MSCs' normal(B),irradiated group1(C1),and irradiated group2(C2),each group had 5 rats.After transplantation the animals were sacrificed on 3rd,5th,7th,14th day respectively.Biochemistry methods were used to detect the serum levels of creatinine(Cr),blood urea nitrogen(BUN) and the quantitation of urine protein per 24 h.Fluorescence microscope was used to identify transplanted MSCs in the renal tissues.Results Compared with group A,the serum of Cr,BUN and the quantitation of urine protein per 24 h in group C hadn't significant difference on the 3rd,5th,7th,14th day(P0.05).In group C,the exogenous MSCs migrated in the renal especially in the renal tubules after five days,and which had nothing to do with the infusion's mode.Conclusion The exogenous MSCs migrate and repopulate in the normal renal tissues,especially in the renal tubules.
