摘要
目的淫羊藿苷对体外培养的人外周血内皮祖细胞(EPCs)增殖的影响。方法采用密度梯度离心法分离获得人外周血单个核细胞,用添加VEGF、FGF的M199培养液培养7d后,经激光共聚焦显微镜、流式细胞仪鉴定为EPCs。用不同浓度药物干预培养EPCs,分别作用不同时间(24、48、72h)后,倒置相差显微镜下观察EPCs生长情况,四唑蓝比色试验(MTT法)测定EPCs的吸光度值。用西药西辛伐他汀作为对照组。结果与对照组比较,淫羊藿苷高、中、低浓度组均改善了EPCs增殖功能(P<0.05),且呈一定的量效、时效关系,在中浓度最为显著(P<0.05)能明显促进增殖能力;在同一浓度组中,随时间延长淫羊藿苷对EPCs的促进作用逐渐增强,于72h达到高峰,且中药中剂量组对EPCs的促进作用优于西药组辛伐他汀组。结论淫羊藿苷能促进EPCs增殖,从而刺激缺血组织血管新生和再生,可能是其防治心脑血管及周围血管疾病的机制之一。
Objective To investigate the effects of Icariine on proliferation of endothelial progenitor cells (EPCs) derived from peripheral bloodderived. Methods The mononuclear cells were isolated from human peripheral blood by.density gradient centrifugation. The laser scanning confocal microscope and the flow cytometry (FCM) was used to determine the EPCs of human peripheral blood. The activity of EPCs was observed under reverse microscope.MTT method was used to determine the OD value of EPCs. Result Compared with control group, the Icariine decoction contained serum high, medium, low dose group increased the proliferative adility of EPCs, showing a time-and do se-effect relation. The Icariine decoction medium dosage group had the most prominent effect on proliferation(P〈0.05). Among the same Icariine decoction dosage group, The effect increased as time went by and reached peak at 72 h. Conclusion Icariine decoction drug-containing serum can premote proliferation of the endothelial progenitor cells, stimulate angiogenesis and regeneration of ischemic tissue .The results may be parts of the mechanisms of Icariine decoction to prevent and treat cardio-/cerebrovascular and peripheral vascular disease.
