我国东乡族Ghrelin基因Leu7 2 Met多态性与2型糖尿病的关系 被引量:5

摘要 目的明确Ghrelin基因Leu72Met多态性在中国甘肃省东乡族的分布并探讨其与2型糖尿病的关系。方法用PCR-DHPLC技术和测序法检测甘肃省东乡族T2DM患者及正常对照者(NC组)Ghrelin基因Leu72Met多态性。结果 Leu72Met基因型和等位基因频率在T2DM组和NC组中的分布差异无统计学意义(P>0.05);NC组cA+AA型的Fins、HOMA-IR明显高于CC型;T2DM组CC基因型的FPG、血肌酐(SCr)水平明显高于cA+AA型(P均<0.05)。结论 Ghrelin基因Leu72Met多态性可能参与胰岛素抵抗和2型糖尿病的发病机制。 Objective To investigate the relationship of ghrelin gene Leu72Met polymorphism with type 2 diabetes mellitus in Chinese Dongxiang nationality. Methods Patients with T2DM and healthy controls were genotyped by PCR-DHPLC and DNA sequencing. Results There were no significant differences in frequencies of allele and genotype in ghrelin gene polymorphism between type 2 diabetes mellitus and normal control group (both P〈0. 05). In normal control group, the subjects with CA+AA genotype had significantly increased levels of Fins, HOMA-IR as compared with CC genotype (P〈0. 05). In diabetic group, the patients carrying CC genotype had the significantly increased levels of FPG and serum creatinine as compared with CA+AA genotype (P〈0. 05). Conclusions Ghrelin gene Leu72Met polymorphism may have some roles in the etiologies of insulin resistance and type 2 diabetes mellitus.
