摘要
以辽阜一号水培芽茎尖为试材,在1/3MS培养基中添加不同生长调节物质诱导茎尖及腋芽生长,研究沙棘的离体培养技术。结果表明:最佳诱导期为3个月,最佳诱导培养基为1/3MS+6-BA 0.2 mg/L+NAA 0.05 mg/L,茎尖生长最好,褐变率低;继代培养基为1/3MS+6-BA 0.5 mg/L+IBA 0.3 mg/L,诱导生根培养基为1/3MS+NAA 0.2 mg/L+IBA0.5 mg/L。
The studying is micropropagation system on Liao--fu No. 1 (Hippophae rharnnoides L. ) with stem apexes as explants under water culture in vitro. Research axillary buds and stem apexes induction through adding different growth regulating material in 1/3MS medium. The results showed the best time of inoculation was March, the inducing medium that stem apexes gets the best growth and the lowest proportion of browning was 1/3MS+6-BA 0. 2 mg/L+NAA 0. 05 mg/L, the multiplication medium was 1/3MS+BA 0. 5 mg/L+IBA 0. 3 mg/L, the rooting medium was 1/3MS+NAA 0. 2 mg/L+IBA 0. 5 mg/L.
