摘要
目的:探讨复健片对大脑中动脉闭塞(MCAO)模型大鼠脑组织P-4502C11mRNA表达的影响。方法:将72只雄性Wistar大鼠随机分为假手术组、模型组和药物组,每组24只,采用电凝大脑中动脉手术造模。自术后1周末始,药物组给予复健片水溶液灌胃,其余两组给予等量生理盐水。在每个观察时点(1周、2周、3周末),各组随机抽取8只大鼠,采用激光多普勒血流仪测定梗死侧额叶局部脑血流量(rCBF),用RT-PCR技术测定梗死额叶周围皮质P-4502C11mRNA的表达。结果:造模结束1周后,与假手术组比较,模型组与药物组梗死侧额叶皮质rCBF、P-4502C11mRNA表达量均显著降低(P<0.01);而治疗1周、2周后,药物组与同期模型组比较,梗死侧额叶皮质的rCBF均有所增加(均为P<0.05),同时,P-4502C11mRNA表达量亦显著增强(分别为P<0.05,P<0.01)。结论:复健片可上调MCAO大鼠受损侧脑组织中原本因缺血而下调的P-4502C11mRNA表达,增强星形胶质细胞释放环氧-二十碳三烯酸(EETs)的能力,这可能是本药改善受损脑组织血流灌注的重要机制之一。
To investigate effect of Fujian tablets on expression of P-4502CllmRNA in middle cerebral artery occlusion(MCAO) rats. Methods:72 male Wistar rats were randomly divided into three groups(n=24 in each group):sham operation,model and drug groups. Middle cerebral artery was occluded by electrocoagulation to make MCAO model. From the end of 1 week after operation,the solution of Fujian tablets was given to drug group by garage method and equal dose of normal salines were given to other two groups. At three different time(the end of one week,two weeks or three weeks after operation),8 rats were selected randomly from each group. LDF was used to measure regional cerebral blood flow(rCBF) of ipsilateral frontal lobes,RT-PCR was used to determine the expression of P-4502CllmRNA in ipsilateral frontal lobes. Results:At the end of 1 week after operation,compared with sham operation group,both rCBF and the expression of P-4502CllmRNA decreased remarkably in model and drug groups(P〈0.01). At the end of two weeks or three weeks after operation,rCBF of drug group increased compared with that of model group(P〈0.05) and the expression of P-4502CllmRNA also increased compared with that of model group at the same time(P〈0.05 and P〈0.01 respectively). Conclusions:Fujian tablets can upregulate the expression of P-4502CllmRNA in MCAO rats, to improve the impaired capacity of astrocytes to secrete EETs induced by cerebral ischemia,which can help increase rCBF of ipsilateral frontal lobes.
