摘要
目的探讨鼠双微基因2(MDM2)和多肿瘤抑制基因(P16)在大肠癌中的表达及临床意义。方法采用免疫组织化学链霉菌抗生物素蛋白—过氧化物酶(S-P)法,对67例大肠癌、30例癌旁组织、20例腺瘤组织,20例正常黏膜的MDM2和P16基因蛋白进行检测。结果MDM2在大肠癌组织中的阳性表达率为71.7%(48/67),明显高于结肠腺瘤组织、癌旁组织及正常黏膜,差异有显著性意义(P<0.05)。P16在大肠癌组织中的阳性表达率为38.8%(26/67),与结肠腺瘤组织的75%、癌旁组织的83.3%及正常黏膜组织的90%比较,差异均有显著性意义(P<0.05)。大肠癌组织中,MDM2在不同分化程度中表达差异无显著性意义(P>0.05)。P16在不同分化程度中表达差异有显著性意义(P<0.05),MDM2和P16在不同Dukes分期中有无淋巴转移之间表达,差异有显著性(P<0.05)。MDM2与P16基因蛋白,在不同年龄组、不同性别组中,表达差异无显著性(P>0.05)。在大肠癌中,MDM2蛋白与P16蛋白表达无明显相关性(P>0.05)。结论MDM2和P16的异常表达,与大肠癌的发生发展有关。联合检测MDM2和P16,对大肠癌的临床诊断、治疗和判断预后,有实际意义。
Objective To study expression of murine double minute gene 2(MDM2) and multiple tumor suppressor(P16) gene in colorectal cancers and its clinical significance.Methods The expression of MDM2 and P16 in 67 specimens of colorectal carcinoma,30 specimens of lateral tissue of carcinoma,20 specimens of colonic adenoma,and 20 specimens of normal colonic tissue was detected by S-P immunohistochemistry.Results In the tissues of colorectal carcinoma,the positive expression rate of MDM2 was 71.7%(48/67).The positive expression rates of MDM2 in the tissues of colorectal carcinoma was much higher than that in lateral tissue of carcinoma,colonic adenoma and the normal colonic tissue.there was significant different(P〈0.05).In the tissue of colorectal carcinoma,the positive expression rates of P16 was 38.8%(26/67),the positive expression rate of P16 in the tissues of colorectal carcinoma was much lower than in colonic adenoma which rate was 75%,in carcinoma which rate was 83.3%,in the normal colonic tissue whi h rate was 90%.there has significant different(P〈0.05).The positive expression rate of MDM2 in variantly differentiated colorectal carcinoma had no significant different(P〉0.05).The positive expression rate of P16 in variantly differentiated colorectal carcinoma had significant difference(P〈0.05).There were significant differences among colonic adenoma in different Dukes staging group,associated with lymphatic metastasis and without lymphatic metastasis(P〈0.05).There were insignificant difference among colonic adenoma in different age group,different gender group(P〉0.05).In the tissues of colorectal carcinoma,the positive expression of MDM2 and P16 had no obvious correlation(P〉0.05).Conclusion The variation of MDM2 and P16 expression in colorectal carcinoma contribute to the occurrence and development of the carcinoma.United detection of MDM2 and P16 could be an important index to treatment,diagnose and prognosis for colorectal carcinoma.
