血浆CK-MM_3单克隆抗体免疫抑制测定法的建立及其临床应用

The establishment and clinical application of McAb inbibition immunoassay of plasma CK-MM_3

人心肌经粗提，离子交换层析，纯化CK-MM抗原。该抗原经杂交瘤技术，制备抗CK-MM单克隆抗体。此抗体特异性抑制血浆CK-MM3。通过免疫抑制法计算抑制活性与CK总活性之比，该比值为心肌梗塞早期诊断指标。此法操作简便、快速、可用于自动分析仪。

CK-MM was purified from human heart. The procedure included: rough fractionation then the CK-MMwas purified by ion exchange chrornatography. The anti CK-MM monoclonal antiboby were obtained bytechnique of cell hybridization. This antibody specially inhibited CK-MM3 in plasma. The specific value (inhibition CK activity) was measured in plasma. This method was used for diagnosing and monitoring acutemyocardial infarction during its early phase and it was easy and rapid to operate.
