摘要
为了探讨胰岛细胞移植的最适宜部位,利用四氧嘧啶诱发的糖尿病鼠,以经门静脉肝内移植为实验组,腹腔内和肌肉内移植为对照组,移植用胶原酶消化提取的乳鼠胰岛细胞.结果,实验组治愈率为60%,与对照组相比有有显著性差异.移植后第8周,未形成肝门静脉高压症,肝脏苏木素伊红染色切片上,未见病理变化,再经胰岛素抗体过氧化物酶抗过氧化物酶法证实肝脏内有散在的胰岛细胞分泌的胰岛素颗粒.实验结果认为,肝脏是特殊免疫器官,排斥反应小,肝内移植易于诱发免疫耐受或免疫无反应性,其特殊的血运和生理功能是胰岛细胞移植的最佳部位.
It has been proved that Wistar rats with diabetes induced by alloxan can be treated with transplantation of new born rats'islet cells. In order to find out the most suitable site for the transplantation of islet cells, the diabetic model rats in this experiment were divided into three groups, one experimental group and two control groups. Intrahepatic transplantation of islet cells was performed in the experimental group while intraperitoneal and intramuscular transplantations of islet cells were respectively performed in the centrol groups. These new born rats' islet cells were extracted from new born rats' pancreases digested by collagenase. The results show that the cure rate is 60% in the experimental group and is significantly different compared with that in the control groups After 8 weeks of the intrahepatic transplantation, no portal vein hypertension was found and no pathological changes were seen in the hepatic sections stained with H-E staining method. The hepatic sections were proved being scattered with insulin granules secreted by islet cells by means of insulin antibody PAP method. The results suggest that the liver is an unique immune organ with minor reject reactions and intrahepatic islet cell transplantation readily induces immunotolerance or immunologic unresponsiveness. Therefore, the liver is the most suitable site for transplantation of islet cells owing to its' unique blood circulation and physiological functions.
