黔产竹节参总皂苷含量测定条件的优化

摘要 为确定香草醛-硫酸-甲醇分光光度法测定黔产竹节参总皂苷含量的优化条件,利用酸性条件下,香草醛与人参皂苷生成有色化合物,进行比色测定,通过正交试验对影响比色反应的因素进行优化。结果显示:有色化合物在496nm有最大吸收,优化条件下制备的总皂苷回归方程为:A=0.0012+0.004716C,相关系数r=0.9991,线性范围为0.007~0.035mg/mL,平均加标回收率为:99.15%,RSD=0.17%(n=5)。表明:该条件下测定黔产竹节参总皂苷含量,结果准确、可靠,重复性好。 The orthogonal test was used for optimizing factors affecting colorimetric reaction of the chemical compound of vanillin and saponin from Panax japonicus produced in Guizhou. The results showed that the maximum absorption of the colored compound was at 496 nm, the regression equation for preparation of total saponin was A: 0.001 2 + 0. 004 716C and its correlated coefficient, linear range, mean recovery and RSD were 0. 9991, 0. 007-0. 035mg/mL, 99. 15% and 0. 17% (n= 5) respectively. Therefore, the vanillin-sulfuric acid-methanol spectrophotometer method for determining total saponin content has accurate, reliable and good repeatable advantages.
