恶性疟原虫对苯芴醇敏感性现场体外测定 被引量:3

摘要 为探索恶性疟原虫对新抗疟药苯芴醇的敏感性现场体外测定方法,提供敏感性数据,1997年8~10月,应用Rieckmann体外微量测定法原理制作苯芴醇测定板,并在云南南部恶性疟流行现场测定了恶性疟原虫对该药的敏感性,重复2次测定的成功率分别为73.8%(31/42)和73.2%(30/41);ID50依次为150nmol/L及130nmol/L;ID95为740nmol/L及760nmol/L;平均抑制裂殖体形成的浓度为422nmol/L及423nmol/L。测定成功率与氯喹板相同,疟原虫在各浓度药井中的抑制率亦同氯喹一样,随药物浓度的增高而依次上升,而裂殖体形成的例数则依次减少。提示本文研制的苯芴醇测定板药性稳定,重复性好,测定结果可靠,可用于恶性疟原虫对该药敏感性体外测定。 To explore methods for testing sensitivity of Plasmodium falciparum to benflumetolum in vitro , and provide the basic data of sensitivity to the drug, a micro test was made and tested in Aug. to Oct. 1997. As following Rieckmann′s in vitro a micro technique, benflumetolum testing plates were coated and response of P. falciparum was assayed with them in the field. The repeated testing(two times) successful rates were 73.8%(31/42) and 73.2%(30/41), ID 50 150 nmol/L and 130 nmol/L, ID 95 740 noml/L and 760 nmol/L and the mean concentration for compete inhibition of schizont formation 422 nmol/L and 423 nmol/L, respectively. In the control of chloroquine testing plates, there were similar results. The higher the drug concentration, the higher the mean schizont inhibited rates and the fewer the munber of case whose blood formed schizont. The results suggested that benflumetolum coated plates were stable, had repetitive and reliable testing results, and could be used for the assay of response of P. falciparum to benflumetolum in the field.
