益气养阴、消癥通络中药对大鼠肾小球系膜细胞分泌细胞外基质的影响

Effect of herb of supplement qi and nourish yin eliminating mass and dredge nels and collaterals on the mesangial cells secreting extracellular matrix

目的探讨益气养阴、消癥通络中药药物血清对大鼠系膜细胞细胞外基质(ECM)不同成分的抑制作用。方法以体外培养大鼠肾小球系膜细胞及血清药理学方法为基础,采用酶联免疫吸附试验检测ECM中纤维连接蛋白(FN)、层黏连蛋白(LN)、Ⅳ型胶原(ColIV)的含量。结果FN、ColⅣ的含量均增加,而益气养阴、消癥通络中药对其具有明显抑制作用。结论益气养阴、消癥通络药物能够减少ECM的积聚,可能是其防治肾小球硬化的机制之一。

Objective To investigate the inhibitory effect of herb of supplement qi and nourish yin eliminating mass and dredge nels and collaterals on the mesangial cells （MCs） secreting extracellular matrix （ECM）.Methods Based on the method of culturing MCs in vitro and serum pharmacology,the contents of fibronectin （FN）,laminin （LN） and collagen Ⅳ （Col Ⅳ） in ECM were detected by ELISA.Results The contents of FN,LN,Col Ⅳ in ECM were all increased,while inhibited by herb of supplement qi and nourish yin eliminating mass and dredge nels and collaterals significantly.Conclusions Herb of supplement qi and nourish yin eliminating mass and dredge nels and collaterals can decrease the accumulation of ECM,which may be one of the mechanisms of preventing glomerulosclerosis.
