地涌金莲的离体培养与快速繁殖技术研究 被引量:6

摘要 [目的]研究地涌金莲(Musella lasiocarpa)的离体培养与快速繁殖技术。[方法]以地涌金莲吸芽为外植体,诱导不定芽分化、增殖和生根。[结果]适宜不定芽增殖的培养基为改良MS+6-BA3.0mg/L+NAA0.3mg/L+AC2.0g/L+Vc0.1g/L+蔗糖30.0g/L+琼脂8.0g/L,增殖系数为4.89;适宜生根的培养基为1/4改良MS+IBA0.5mg/L+蔗糖20.0g/L+琼脂8.0g/L,生根率达100.00%,平均生根数4.60条,平均根长3.40cm;幼苗移栽成活率达91%。[结论]为大面积推广应用地涌金莲奠定了基础。 [Objective] The research aimed to study the in vitro culture and rapid propagation technology of Musella lasiocarpa. [Method] The surculus sucker from Musella lasiocarpa were used as explants to induce buds and rooting in different culture media. [Result] For its buds propagation,the optimum culture medium was MS+6-BA 3.0 mg/L+NAA 0.3 mg/L+AC 2.0 g/L+Vc 0.1 g/L+ sugar 30 g/L+ agar 8 g/L,with reproduction coefficient 4.89. The optimum culture medium for rooting was 1/4 MS+IBA 0.5 mg/L + sugar 20 g/L+ agar 8 g/L,where the rooting rate could achieve to 100.00%,with its average rooting number 4.60 and average rooting length 3.40 cm. The transplant survival percent was 91%. [Conclusion] The study can provide the basis for applying Musella lasiocarpa in large scale.
