摘要 目的:研究宫颈脱落细胞中人端粒酶RNA组分(hTERC)基因的表达,探讨其在宫颈上皮内瘤变(CIN)及鳞状上皮细胞癌(SCC)诊断中的价值。方法:以2007年12月~2008年12月在中日友好医院行子宫颈癌筛查的138例妇女为研究对象,进行宫颈液基细胞学及组织病理学的检测,以组织病理学不同级别分组,荧光原位杂交方法检测其hTERC基因的表达。结果:病理学确诊为正常、CINⅠ、CINⅡ、CINⅢ及子宫颈癌例数分别为20、28、31、34、25例,宫颈脱落细胞hTERC基因的扩增在不同组织病理级别中存在差别:hTERC基因扩增随细胞学级别的升高而升高,随组织病理学级别的增加而升高。结论:hTERC基因的扩增与宫颈细胞学和组织病理学级别密切相关,其扩增率随宫颈病变程度的加重而增加;检测hTERC基因的扩增,可为判断是否发展为宫颈高度病变提供依据。 Objective:To investigate amplification of the human telomerase RNA component (hTERC)gene in the cervical exfoliated cells and to explore its value in diagnosis and treatment of cervical intraepithelial neoplasia (CIN)and malignancy.Methods:One hundred and thirty eight women were examined with liquid-based cytology and hTERC gene detection using fluorescence in situ hybridization (FISH) in China-Japan Friendship Hospital.Results:The case number of normal,CINI, 11,11I ,cervical cancer were 20,28,31,34,25 respectively. There were significant difference of hTERC gene amplification among the groups with different cytological and histological lesion.In cytological groups,there was a different increase of hTERC gene amplification in patients with high-grade squamous cervical intraepithelial lesion and above lesion.In histological groups,there is a dif- ferentl increase of hTERC gene amplification in patients with CINII and severity.Conclusion:There is a tight correlation between amplification of hTERC and the histological and cytological lesion,it increased progressively along with the severity of cytological and histological grades.The hTERC amplified or not maybe as a prognostic indicator to judge lesion's severity.
