摘要
目的:通过观察葶苈生脉方对充血性心力衰竭(CHF)大鼠心肌纤维化及转化生长因子β1(TGF-β1)表达的影响,探讨其治疗CHF的作用机制。方法:60只雄性Wistar大鼠随机分为假手术组、模型对照组、葶苈生脉方高剂量组、葶苈生脉方低剂量组、阳性药心宝丸对照组。采用腹主动脉缩窄法复制CHF大鼠模型,喂养4周后分别给予蒸馏水或药物,8周后检测血流动力学参数,用MASSON染色法观察左室心肌胶原形态,图像分析测量胶原容积分数(CVF),免疫组化法检测心室肌中TGF-β1的表达。结果:与假手术组比较,模型对照组CVF明显升高(P<0.01);与模型对照组比较,各治疗组CVF则显著降低(P<0.01);与假手术组比较,模型对照组TGF-β1表达增强(P<0.01);与模型对照组比较,各治疗组TGF-β1表达则减弱(P<0.01)。结论:葶苈生脉方能明显改善心室重构和心肌纤维化,改善心功能,此作用可能与影响TGF-β1表达有关。
Objective: To explore the mechanism of congestive heart failure (CHF) by observing the effects of Tingli Shengmai Decoction on myocardial fibrosis and expression of TGF-β1 of rats with CHF. Methods: The CHF animal models were duplicated by the abdomen arteriarctia method, and 60 male wistar rats were randomly divided into the sham-operation group, the model group, the high-dose of Tingli Shengmai Decoction group, the low-dose of Tingli Shengmai Decoction group and the positive medicine (Xinbaowan) control group. After 4 weeks common fed, every group rats were given a certain dose of distilled water or medicine. After 8 weeks, hemodynamic parameters were detected, MASSON staining was used in the study of collagen type in left ventricular interstitial tissue, collagen volume fraction (CVF) were measured by image analysis, and expression of TGF-β1 in myocardium were evaluated by immunohistochemical staining. Results: Compared with the sham-operated group, CVF of model control group increased significantly (P0.01); Compared with the model control group, CVF of treatment groups decreased (P0.01). Compared with the sham-operated group, TGF-β1 expression of model control group increased (P0.01); Compared with the model control group, TGF-β1 expression of treatment groups was reduced (P0.01). Conclusion: Tingli Shengmai Decoction can obviously prevent myocardial remodeling and myocardial fibrosis, improve cardiac function, which may be related with the impact of TGF-β1 expression.
