摘要
目的:参照环介导等温扩增技术(Loop-mediated isothermal amplification,LAMP)的基本原理、特点,并应用于假结核耶尔森菌(Yersinia pseudotuberculosis,Y.pseud)的检测。方法:应用LAMP技术,针对Y.pseud的inv基因设计合成6条引物(2条内引物,2条外引物及2条环引物),对12株耶尔森菌进行检测,并将其结果与传统结果相比较。结果:所检测的菌株样本中3株Y.pseud经LAMP检测均显绿色为阳性,而其余致病性和非致病性的Y.ent均显橙色为阴性。结论:LAMP方法快速、特异性强且操作简便,极适宜在基层实验室应用。
Objective:To apply Loop-mediated isothermal amplification(LAMP) for the detection of Yersinia pseudotuberculosis.Methods:Using the technology of LAMP to detect tewlve Yersinia strains and the result was compared with the traditional result.Six primers were designed and synthesized according to inv gene of Y.pseud(tow inter primers,tow outer primers,tow ring primers).Results:Three strains of Y.pseud by detection of LAMP were significantly positive for the green and the remaining Y.ent strains with pathogenic and non-pathogenic were significantly negative for the orange.Conclusion:LAMP method is fast,strong and easy to operate,which is specifically appropriate for the grassroot level laboratories.
