三聚氰胺单克隆抗体的制备与鉴定

Preparation and Identification of Monoclonal Antibodies Against Melamine

本实验利用三聚氰胺类似物与血蓝蛋白、卵清蛋白偶联制备免疫抗原和包被抗原,免疫Balb/c小鼠,采用B细胞杂交瘤技术,经免疫、融合、筛选和克隆等,得到了抗三聚氰胺单克隆抗体。该抗体亚类为IgG1,制备的单克隆抗体腹水效果评价达10-7,与其他三聚氰胺结构类似物无交叉反应。三聚氰胺单克隆抗体的获得为进一步研发三聚氰胺药物残留检测试剂盒的研制奠定了基础。

A monoclonal antibody（McAb） against Melamine was prepared for measuring Melamine residuals and for an enzyme linked immuno-sorbent assay（ELISA） reagent kit. Balb/c mice were immunized intraperitoneally with Melamine coupled with KLH and OVA.Hybridoma cell lines secreting McAbs were obtained using routine hybridoma technology and ELISA screening.The McAbs subtype was IgG1.The ascite titer was 10^-7.The McAbs had no cross-reaction with some analogues of Melamine.The results show that the specific MELMcAbs are developed and these McAbs may be useful in the development of detection assay.
