磷酸肌酸钠对小鼠移植性S180肉瘤生长的影响 被引量:2

摘要 目的:观察不同剂量磷酸肌酸钠(CP)对移植性小鼠S180肉瘤增殖及凋亡的影响,并初步探讨其可能的作用机制。方法:皮下注射S180肉瘤细胞建立动物实体瘤模型,将小鼠随机分为生理盐水对照组和CP4mg/d、8mg/d、16mg/d给药组,连续腹腔用药7d,取出瘤组织后应用流式细胞分析技术(FCM)检测S180肉瘤组织凋亡率及PCNA蛋白表达水平,并应用免疫组织化学法检测S180肉瘤组织中Bcl-2和Bax蛋白的表达水平。结果:给药组平均瘤重较对照组减轻,凋亡率无明显变化。各干预组PCNA蛋白的表达均低于对照组,其中CP16mg/d组与对照组有显著差异,Bcl-2/Bax的比值在对照组最高,各组间的差别无统计学意义。结论:治疗量的磷酸肌酸钠[CP(4~8)mg/d]应用于移植性小鼠S180肉瘤不影响肿瘤的增殖及凋亡;当高剂量(16mg/d)应用时可抑制肿瘤的生长。 Objective:Creatine phosphate sodium(CP)is a medicine used to protect myocardium in many kinds of heart disease.But there were a lot of debate on whether CP use to the people of cancer to protect themyocardium injure because of the chemotherapy.To study the growth and apoptosis influence of CP on expression of PCNA protein、Bcl-2 and Bax on S180 transplant tumor.Methods:Mouse solid tumor model was established by subcutaneous injection of S180 cells,mices were divided into four groups randomly :saline control group and low,middle and high creatine phosphate sodium for injection treatment groups,mice were i.p.injected saline or different consistency drug for consecutively 7days,FCM were used to detect the cell apoptosis and the expression of PCNA protein;The expression levels of Bcl-2 and Bax protein were detected with immunohistochemistry method.Results:The average weight of the transplant tumors of the treatment groups were smaller.The change of cell apoptotic rate is not markedly.The expression of PCNA protein in treatment groups were decreased,between the CP16mg/d group and the control group,the difference is significant.The ratio of Bcl-2 /Bax have no marked change.Conclusion:The therapeutical dose of CP[(4 ~8)mg/d]don't affect the growth and apoptotic of the S180 transplant tumor.The high dose CP(16mg/d)inhibit the growth of S180 transplant tumor.
