实验性变态反应性脑脊髓炎豚鼠外周血单个核细胞分泌IL-4量的研究 被引量:2

摘要 目的:观察急性实验性变态反应性脑脊髓炎(EAE)豚鼠外周血单个核细胞(PBMC)IL-4分泌量,探讨其与EAE发病情况的相关性。方法:将30只雌性豚鼠随机分为正常对照组、EAE模型组和佐剂对照组,每组各10只。采用粗制髓鞘碱性蛋白诱发豚鼠急性EAE。记录上述三组豚鼠发病的潜伏期、进展期及神经功能障碍评分,并采用ELISA法测定PBMC培养上清液中IL-4量的变化。结果:EAE模型组在接种约2周后出现厌食、体重下降,不同程度的肢体瘫痪,且均表现为单病程发病;正常对照组和佐剂对照组未发病。EAE模型组PBMC分泌IL-4量较正常对照组降低;而佐剂对照组与正常对照组差异无统计学意义。EAE模型组豚鼠PBMC分泌IL-4的量与发病的潜伏期呈正相关,与进展期及神经功能障碍评分呈负相关。结论:EAE豚鼠体内存在免疫失衡,IL-4分泌量减少,且与发病的严重程度有相关性。
