摘要
目的从基因水平探讨肿瘤坏死因子(TNF-αR1)及半胱氨酸蛋白10(Caspase-10)在增生性瘢痕组织中的作用,为增生性瘢痕的基因治疗提供理论依据。方法利用逆转录聚合酶链反应(RT-PCT)法分别检测12例增殖期增生性瘢痕组织、12例正常皮肤对照组织中成纤维细胞TNF-αR1 mRNA及Caspase-10 mRNA的表达情况。结果在增生性瘢痕组织中,TNF-αR1 mRNA及Caspase-10 mRNA表达的相对值分别为(1.31±0.27)和(1.19±0.23),均明显低于正常皮肤组织(P<0.05)。结论TNF-αR1及Caspase-10在伤口愈合过程中起着重要的作用,增生性瘢痕的形成可能与组织中TNF-αR1 mRNA及Caspase-10 mRNA表达低下有关。
Objective To explore the role of tumor necrosis alpha receptor 1(TNF-αR1)and Caspase-10 in hypertrophic scar during multiplication period at the gene level.Methods The expression of TNF-αR1 mRNA and Caspase-10 mRNA in 12 cases of hypertrophic scars during multiplication period was tested by semi-quantitative reverse polymerase chain reation(RT-PCR)method and was compared with that of the 12 cases of normal skin.Results The expression rate of TNF-αR1 mRNA and Caspase-10 mRNA in hypertrophic scar were(1.31±0.27)and(1.19±0.23),respectively,which were significantly lower than those of the normal skin(both P〈0.05).Conclusion TNF-αR1 and Caspase-10 may participate in the process of wound healing and that the formation of hypertrophic scar might be due to the down-regulation of TNF-αR1 and Caspase-10.
