摘要
目的探讨碱性成纤维细胞生长因子(bFGF)对骨缺损修复的影响。方法取健康家兔24只,随机分为实验组和对照组,将每只兔右侧桡骨以桡骨隆凸处为中心造成10mm长的完全骨-骨膜缺损。实验组于手术当日及术后第7天、14天、21天往骨缺损处局部注射100μl含游离bFGF50ng的生理盐水,对照组局部注射100μl的生理盐水。术后2周、4周、8周通过大体标本观察、X线摄片、骨密度测定、组织学检查等相关指标检测,分析两组骨缺损处的骨痂生长情况。结果(1)X线检查:术后4周与8周实验组桡骨骨缺损断端出现骨痂并相对生长,骨缺损断端间骨痂逐步增多,骨缺损断端间的距离逐步减小,而对照组骨痂较少。(2)单光子骨密度(BMD)测定:术后4周与8周实验组桡骨断端骨密度高于对照组,差异有统计学意义(P<0.05)。(3)组织学观察:术后4周与8周,实验组新生骨组织的形成量逐步增多,而对照组无明显变化。结论碱性成纤维细胞生长因子在动物体内骨缺损局部应用显示良好的成骨作用,对骨缺损的修复有明显的促进作用。
Objective To explore the effect of local injection of basic fibroblast growth factor(bFGF)on the repair of bone defect.Methods 24 healthy adult rabbits were randomly divided into experimental and control groups,the right side of each rabbit radius were made a complete 10mm long bone-periosteal defect with radius knob as the center,of the experimental group,on the operation day and the 7th day,14th day,21st day after operation,free bFGF 50 ng in 100 μl normal saline was injected into the bone defect,while control group was injected 100 μl of normal saline.The relevant indicators such as gross specimen observation,X-ray examination,bone mineral density(BMD)measured,histological observation,analysis were observed At the 2nd,4th and 8th weeks after operation and compared the callus growth between two groups.Results (1)X-ray examination:at the 4th and 8th weeks after surgery,there was the relative growth of callus on the bone ends of bone defect in experimental group,the callus growth between the bone ends increased significantly,the distance between bone ends smaller,while the control group presented no significant change.(2)single-photon bone mineral density(BMD)measured:at the 4th and 8th weeks after surgery,higher bone mineral density was observed on the stump of bone defects in experimental group than in control group and with statistical significance(P〈0.05).(3)Histological observation:at the 4th and 8th weeks after surgery,the formation of new bone tissue further increased in experimental group,while no significant changes in control group.Conclusion Local administration of bFGF in animals show well osteogenesis effect and promote the repair of bone defect.
