摘要
目的:研究关龙胆乙醇提取物对急性肺损伤的保护作用。方法:NIH雄性小鼠48只,随机均分为空白(等量生理盐水)、模型(等量生理盐水)、醋酸地塞米松(0.005g·kg-1)及关龙胆乙醇提取物高、中、低剂量(10、5、2.5g·kg-1)组。灌胃给药,每天1次,连续5d。末次灌胃后以脂多糖(LPS,2.5mg·kg-1)气管滴注复制急性肺损伤模型。复制模型24h后以酶联免疫法检测血浆前列腺素E2(PGE2)、白介素-10(IL-10)和环氧化酶-2(COX-2)的含量。结果:与模型组比较,关龙胆乙醇提取物高、中、低剂量组血浆PGE2和COX-2水平显著降低(P<0.05或P<0.01),血浆IL-10水平显著升高(P<0.05或P<0.01)。结论:关龙胆乙醇提取物可能通过抑制COX-2表达、降低PGE2生成,并促进IL-10的生成对急性肺损伤产生保护作用。
OBJECTIVE: To investigate the protective effects of the ethanol extract of Radix et Rhizoma Gentianae on acute lung injury (ALI). METHODS: 48 NIH male mice were randomized into blank group (normal saline), model group (normal saline), dexamethasone acetate group (0.005 g.kg^-1), ethanol extract of Radix et Rhizoma Gentianae groups (high-dosage of 10 g. kg^-1, medium dosage of 5 g. kg^-1 and low dosage of 2.5 g. kg^-1). The groups were administered via i.g gtt once a day for five days. ALl model was induced by intratracheal injection of lipopolysaccharide (LPS, 2.5 mg. kg^-1) after last treatment. ELISA method was applied to determine the content of prostaglandin E2 (PGE2), interleukin-10 (IL-10) and cycloxygenase-2(COX-2) at 24 h after modeling. RESULTS: As compared with model group, the content of PGE2 and COX-2 in ethanol extract of Radix et Rhizoma Gentianae groups were decreased significantly (P〈0.05 or P〈0.01) while the content of IL-10 was increased significantly (P〈0.05 or P〈0.01). CONCLUSION: Ethanol extract of Radix et Rhizoma Gentianae protect lung by inhibiting the expression of COX-2, reducing the production of PGE2 and promoting the generation of IL- 10.
