摘要
目的探讨导流杂交基因芯片技术和第二代杂交捕获法检测宫颈液标本中人乳头状瘤病毒(HPV)感染的一致性,评价导流杂交基因芯片技术的临床应用价值。方法采用导流杂交基因芯片技术和第二代杂交捕获法对356例疑似HPV感染的标本进行基因诊断,对检测结果不一致的标本进行DNA测序,根据测序结果进行HPV分型,比较这两种方法对13种高危型HPV检测结果的符合情况。结果导流杂交基因芯片技术检测13种高危型HPV的检出率78.9%(281/356),其中单一感染217例,多重感染64例;第二代杂交捕获法检测结果阳性280例;两种方法检测13种高危型HPV的总符合率92.6%,总Kappa指数(KI)0.891,二者有很好的一致性。结论导流杂交基因芯片技术适合更临床筛查HPV感染及对HPV进行基因分型。
Objective To evaluate the relevance of two detection methods of human papillomavirus virus gene(HPV).Methods A total of 356 women who visited People's Hospital of Henan province were selected,and their cervical scraping samples were typed by HybridMax for 21 types of HPV DNA,and were detected by Hybrid Capture 2(HC-2)for 13 types of high-risk HPV in the meantime.The accordance of two methods were compared.Results The accordance of two methods was good.All Kappa index was 0.891.Conclusion HybridMax was comparable to HC-2 for HPV detection,and could also distinguish HPV genotypes definitely,so it is an ideal method for HPV testing.
