外源性透明质酸在愈合中伤口的生化分析

摘要 目的探讨外源性透明质酸(HA)抑制瘢痕形成的作用及其可能机制。方法健康SD大鼠45只,复制背部创伤模型,随机分成2%HA治疗组(A组)、1%HA治疗组(B组)、PBS对照组(C组),观察创面大体形态、平均愈合时间和残留面积的大小,测定愈合部位组织蛋白、羟脯氨酸(Hyp)、Ⅲ型前胶原(PCⅢ)的含量。结果⑴3组的平均愈合时间为(15.2±2.0)d、(14.2±1.6)d和(13.4±1.4)d,A、B两组与C组比较有显著性差异,3组间比较也有显著性差异(P<0.05)。A、B两组的残留面积与C组比较有显著性差异,3组间比较也有显著性差异(P<0.05)⑵Hyp和PCⅢ含量的测定:A、B两组Hyp含量的均值低于C组(P<0.01);而PCⅢ的含量却高于C组(P<0.01)。结论⑴HA通过对细胞的调控作用,有效地抑制胶原的合成,提高Ⅲ型胶原含量,使创伤愈合过程中瘢痕形成减少。⑵HA在创面愈合中抑制瘢痕形成的生物学作用与其浓度成正比关系。 Objective To evaluate the effects and possible mechanisms of hyaluronic acid to inhibit scar formation.Methods 45 health adult SD rats in a dorsal part trauma model were randomly divided into 2% HA treated-group(A group),1% HA treated-group (B group),and PBS treated-group as a control group (C group).The process of healing was observed,the remained wound areas were measured,and a mean wound healing time were calculated.Furthermore,dynamic analysis were performed on local tissure protein,Hyp and PC Ⅲcontents.Results ⑴The mean value of wound healing time was (15.2±2.0)d,(14.2±1.6)d and (13.4±1.4)d respectively.The remained wound areas of control group were much smaller than that in A,B groups ( P <0.05),and there was marked difference among the three groups( P <0.05).⑵The contents of tissue Hyp statistically were lower in HA-treated groups,while the contents of PC Ⅲ were statistically higher comparing to control group ( P <0.01).Conclusion The study indicated that ⑴the exogenous hyaluronan can effectively inhibit collagan synthesis and elvate tissue PC Ⅲ content.As a result,scar formation was decreased.⑵Hyaluronan efficacy presented a dose-dependent manner.
