摘要
通过对酶解法、热回流法、超声波和微波法提取双孢蘑菇中核酸效果进行了比较,结果表明微波法优于其它方法。最终,确立采用微波破壁提取双孢蘑菇中核酸。经单因素(时间、功率、NaCl%、pH)实验和正交实验,得最佳提取条件为:30min、300W、4%NaCl、pH为11。用定磷法测得双孢蘑菇核酸提取率为2.78%。
Four methods of extracting nucleic acid from Agaricus bisporus by enzymatic, heat reflux, ultrasonic and microwave were compared there.Through the experiment it was found that microwave method is better to break Agaricus bisporus for extracting nucleic acid.So the microwave method was chosen in the end.By means of single factor ( time, power, NaCl%, pH value) experiment and orthogonal test, it got the best extraction condition ;30 min, 300W,4% NaCI, pH value of 11. The nucleic acid content of Agaricus bisporus was 2.78% by phosphate determination method.
